Hamato Yoshi
Finish Challenge Mode as Raphael, Donatello, Michaelangelo, or Leonardo to unlock Hamato Yoshi in versus mode.

Alternate costumes
Simply set the system date to December 25 to see the turtles with a Christmas theme. Also set the system system date to October 31 to see the turtles with a Halloween theme.

Challenge mode
Finish story mode as Raphael, Donatello, Michaelangelo, and Leonardo.

Oroku Saki
Beat Saki as Raphael, Donatello, Michaelangelo, or Leonardo to unlock Oroku Saki in versus mode.

Shredder
Beat Shredder as Raphael, Donatello, Michaelangelo, or Leonardo to unlock Shredder in versus mode.

Casey Jones
Finish level 1 as Raphael to access Casey Jones in versus mode.

Evil Turtlebot
Finish level 3 as Raphael, Donatello, Michaelangelo, or Leonardo to have the Evil Turtlebot in versus mode available.

Hun
Finish level 6 as Michaelangelo to have Hun in versus mode available.

Splinter
Finish Dojo level 3 as Leonardo to have Splinter in versus mode available.
